凝血酶为牛血或猪血中提取的凝血酶原,经激活而得凝血酶的无菌冻干品,为白色或类白色的冻干块状物或粉末。干粉在2~80C储存很稳定,干粉易溶于水,0.9%的氯化钠溶液显浑浊,不溶于有机溶剂。在室温水溶液8h内失活。故须临用时新鲜配制,遇热、称酸、碱、金属等活力降低。。

应用领域
凝血酶为局部止血药,局部应用无副作用。使用时必须直接与创面接触,才能起到止血作用。但是本品严禁注射,不得与酸、碱及重金属配伍。动物试验有机抗原体反就、过敏反应。如出理此症状,应立即停药。局部止血用氯化钠注射液溶解成含50~250单位/ml的溶液或干粉,喷雾或洒于创伤表面。消化道止血,用温开水(≤370)溶解成10~100单位/ml的溶液,品服或局部灌注,根据出血部位及程度适当增减浓度、次数。局部止血药,用于局部出血及消化道出血生化研究
制备方法/合成路线
原料处理、沉淀取猪血液2kg,加入枸橼酸钠(38g/L即3.8%)溶液200ml抗凝,混匀,离习分离,取出上清液血浆,在血浆加入10倍于血浆量的蒸馏水稀释,用300ml乙酸(1%)调节Ph为5.3,待沉淀完全,于离心机中离心5~10min,弃去离心液,收集沉淀,得凝血酶原。动物血液枸橼酸钠→原料处理血浆蒸馏水:HAc;Ph5.3→沉淀凝血酶原分离、沉淀、洗涤取凝血酶原,在25~ 30C0下,加入氯化钠(0.9%)的溶液700ml,使之溶解,再在每升溶液中加入15g氯化钙,充分搅拌10min,使之溶解,冷室置1h,离心分离,取上清液加等量冷冻丙酮,搅拌均匀后,静置过夜。离心分离,沉淀再加冷冻丙酮研细,冷室放置2~ 3d,过滤,得沉淀后用乙醚洗涤,真空干燥,即得粗制凝血酶,约10g。凝血酶原NaCl;CaCl2;丙酮→分离;沉淀沉淀乙醚→洗涤粗制凝血酶除杂质、沉淀、干燥在粗制凝血酶中加入200ml氯化钠液(9g/L),使之溶解,溶解后00C放置6h,过滤,取沉淀再加150ml氯化钠液(9g/L)重复以上操作1次,合并两次滤液。用乙酸(1%)调节Ph至5.5。离心分离,沉淀弃去,取上清液加入2倍于上清液量的冰冻丙酮,静置2h,待沉淀完全,离心分离,沉淀用冷冻丙酮研细,放置24h,滤去溶液。沉淀分别用无水乙醇、乙醚洗涤,真空干燥,即得精致凝血酶,约0.4g。粗制凝血酶NaCl;HAc;^0<0C;pH5.5→滤液丙酮→沉淀干燥精制成品
